Is it possible to play Wiim Ultra USB disk files, from the Alexa device speaker? What is the procedure?

No, Alexa is only aware of cloud supported services such as Amazon music etc and isn’t aware of any local media whether that be on the Ultra’s usb drive, your phone, PC etc
